SLU-PP-332
SLU-PP-332 is a synthetic small molecule that acts as an agonist of the estrogen-related receptors (ERRα, ERRβ and ERRγ), with its strongest activity at ERRα. It was developed at Saint Louis University as a tool for a receptor family that had few usable agonists. Chemically it is an aryl hydrazone built from a 4-hydroxybenzohydrazide and a naphthaldehyde. Rogue Compounds supplies it in capsule form.

The receptor family
Despite the name, ERRs do not bind estrogen. They are orphan nuclear receptors that are constitutively active and work with the coactivators PGC-1α and PGC-1β to control genes for mitochondrial biogenesis, fatty-acid oxidation and oxidative phosphorylation. ERRα and ERRγ are highly expressed in tissues with high energy demand, such as heart, skeletal muscle, kidney and brown fat. For years most ERR pharmacology relied on inverse agonists, which made a synthetic agonist useful for asking what happens when the pathway is pushed upward.
What researchers study
- Receptor pharmacology: cell-based transactivation assays comparing activity at the three ERR isoforms.
- Muscle gene programs: C2C12 myocytes and mouse skeletal muscle, looking at ERRα-dependent oxidative and mitochondrial gene expression and fiber-type markers, and how they compare with the response to aerobic exercise.
- Metabolic models: diet-induced and genetically obese mouse models, measuring energy expenditure, fatty-acid oxidation and tissue gene expression.
The published literature is preclinical and comes largely from a single research group. Findings in mice do not establish effects in humans.
Regulatory status
SLU-PP-332 is not approved by the FDA, has not been tested in published human studies, and is not approved for human use. It is a research reagent only.
Quality & Documentation
Every batch of SLU-PP-332 is made in small runs for Rogue Compounds and tested by an independent U.S. laboratory for identity (mass spectrometry) and purity (HPLC). The batch Certificate of Analysis is posted on this page and in our COA Library, so the pouch in your lab can be matched to its report.
