Background on the compound

5-Amino-1MQ was identified in a structure-activity study that screened a series of quinolinium analogs against NNMT, the enzyme that methylates nicotinamide using S-adenosylmethionine. Researchers compared how substitutions around the quinolinium ring changed binding in the enzyme’s nicotinamide pocket, and 5-amino-substituted analogs were among the more potent inhibitors in that series.

Follow-up preclinical work examined the compound in cultured adipocytes and in diet-induced obese mice, measuring 1-methylnicotinamide, intracellular NAD⁺ and SAM, and adipose tissue readouts. A separate rodent study looked at muscle stem cells in aged mice, asking whether NNMT inhibition changes their activity in injury models. All published work is preclinical.
